Main function

TRPM8 helps the body detect cool and cold temperatures and is activated by cooling compounds such as menthol. It is mainly found in sensory nerves and contributes to normal cooling sensations, cold pain and the pain-relieving effect that mild cooling can sometimes produce.

Positive and negative describe interaction direction, not whether an effect is good or bad. Activating a target can have unwanted effects, while reducing or blocking it can sometimes have beneficial effects.

Positive interaction

Increased activity

Increased TRPM8 activity increases cooling sensations and sensitivity to cold. Moderate activation can reduce some types of pain, while excessive activation may increase cold-related pain and hypersensitivity.

Negative interaction

Reduced or blocked activity

Reduced or blocked TRPM8 activity decreases sensitivity to cold and can reduce cold-related pain and hypersensitivity. It may also reduce the pain-relieving effects normally produced by mild cooling.
